چکیده

Introduction The concentration of serum creatinine is widely used as an indicator of renal function. Sarcosine oxidase (SO) has been used in the enzymatic determination of creatinine in the clinical laboratory. SO has been isolated from various bacterial strains. It was first purified from Corynebacterium sp. U-96 (SO-U96) and shown to be a heterotetrameric enzyme. SO-U96 is a complex, bifunctional enzyme that catalyzes both sarcosine oxidation and the synthesis of 5,10methylenetetrahydrofolate. SO-U96 contains four

Analysis of water channels by molecular dynamics simulation of heterotetrameric sarcosine oxidase

A precise 100-ns molecular dynamics simulation in aquo was performed for the heterotetrameric sarcosine oxidase bound with a substrate analogue, dimethylglycine. The spatial region including the protein was divided into small rectangular cells. The average number of the water molecules locating within each cell was calculated based on the simulation trajectory. The clusters of the cells filled ...

A stable three-enzyme creatinine biosensor. 1. Impact of structure, function and environment on PEGylated and immobilized sarcosine oxidase.

The determination of creatinine levels in biological fluids is an increasingly important clinical requirement. Amperometric biosensors have been developed based on a three-enzyme system which converts creatinine to amperometrically measurable hydrogen peroxide. The development of the amperometric creatinine biosensor has been slow due the complexity of the three-enzyme system. This paper, the f...

Oxygen Pathways and Allostery in Monomeric Sarcosine Oxidase via Single-Sweep Free-Energy Reconstruction

Monomeric sarcosine oxidase (MSOX) is a flavoprotein D-amino acid oxidase with reported sarcosine and oxygen activation sites on the re and si faces of the flavin ring, respectively. O2 transport routes to the catalytic interior are not well understood and are difficult to ascertain solely from MSOX crystal structures. A composite free-energy method known as single-sweep is used to map and ther...

Structure-function correlation in glycine oxidase from Bacillus subtilis.

Structure-function relationships of the flavoprotein glycine oxidase (GO), which was recently proposed as the first enzyme in the biosynthesis of thiamine in Bacillus subtilis, has been investigated by a combination of structural and functional studies. The structure of the GO-glycolate complex was determined at 1.8 A, a resolution at which a sketch of the residues involved in FAD binding and i...

The cofactor requirements of sarcosine oxidase.

It has been established that the enzyme, sarcosine oxidase, found in the mitochondria of rat liver and elsewhere (I), catalyzes the following over-all reaction (2, 3) : sarcosine + $02 = glycine + serine + HCHO. The cofactor requirements of this reaction have not been established, although Mackenzie (1) has mentioned unpublished results which seemed to indicate that a folic acid deficiency in r...
